Human RTN4RL1/NgR3/NGRH2/NGRL2 Gene, Full-length cDNA in Lentivector, Endotoxin-free DNA
RTN4RL1 (Reticulon-4 receptor-like 1) ia a GPI-anchored membrane protein that belongs to the Nogo receptor (NgR) family. It has a 396 amino acid (aa) extracellular region with 8 LRR (leucine-rich) repeats. RTN4RL1 is predominantly expressed in brain with lower levels in kidney, lung, mammary gland, placenta, salivary gland, skeletal muscle and spleen. RTN4RL1 may function similar to RTN4R that act as a receptor for reticulon 4 (RTN4), oligodendrocyte myelin glycoprotein (OMG) and myelin-associated glycoprotein (MAG). RTN4R mediates axonal growth inhibition and may play a role in regulating axonal regeneration and plasticity in the adult central nervous system (CNS). RTN4R mediates the activation of Rho and downstream reorganization of the actin cytoskeleton. RTN4R acts in conjunction with RTN4 and LINGO1 (Leucine-rich repeat and immunoglobulin-like domain-containing nogo receptor-interacting protein 1) in regulating neuronal precursor cell motility during cortical development. RTN4RL1 may thus play a role in regulating axonal regeneration and plasticity in the adult central nervous system.Localized to the surface of neurons, including axons. RTN4RL1 may undergoe zinc metalloproteinase-mediated ectodomain shedding in neuroblastoma cells; where it is released as a full-length ectodomain and an N-terminal fragment containing the leucine-rich repeat (LRR) region.
Gene Symbol: RTN4RL1 ;NgR3; NGRH2; NGRL2; R4RL1
NCBI Gene ID: 146760
Uniprot Entry: Q86UN2
Construct Details: Full length human RTN4RL1 gene is subcloned into the Lentiviral expression vector pLTC with an upstream CMV promoter and with or without a selection marker, which can be used for both transient and stable expression in mammalian cells. It can be co-transfected with the LentiPAK DNA mix (SKU# LP-001) into HEK293 cells to produce high titer lentiviral particles. Multiple choices of a stable antibiotic selection marker are available.
Vector Type: pLTC or pLTC-IRES-Marker (lentiviral expression vector containing a heterologous CMV promoter, see the vector map above)
Formulation: Supplied with 10 μg of endotoxin-free plasmid DNA at 200 ng/μl in sterile TE buffer (pH8.0)
Gene Insert Size: 1326 (bp)

Primers: Gene insert coding sequence can be confirmed by following primers:
(a) forward (or 5'-end) primer: 5’-CAAATGGGCGGTAGGCGTG-3’;
(b) reverse (or 3'-end) primer: 5’-CCAGGTTTCCGGGCCCTCAC-3’
Note: The sequence may vary due to the naturally occurring variants or mutations, such as single nucleotide polymorphism, or the artifact during PCR amplification. High fidelity PCR systems should always be used.
