Human CD2/T11/SRBC/LFA2 Gene, Full-length cDNA Clone in Lentivector, Endotoxin-free DNA
T-cell surface antigen CD2, also known as erythrocyte receptor, rosette receptor, T11/Leu-5, SRBC and LFA-2, is a single-pass type I transmembrane protein of the CD2 family of the immunoglobulin (Ig) superfamily. CD2 contains one Ig-like V-type (IgV) and one Ig-like C2-type (IgC2) domain in the extracellular region. CD2 is one of the earliest T-cell markers, being present on more than 95% of thymocytes; CD2 is also found on some natural killer (NK) cells but not on B lymphocytes. The CD2 family of receptors are evolutionarily conserved and regulate T cell- and NK cell-mediated cytotoxic activity and inflammatory cytokine production when engaged by ligands. Monoclonal antibodies directed against CD2 inhibit the formation of rosettes with sheep erythrocytes, indicating that CD2 is the erythrocyte receptor or is closely associated with it. CD2 interacts with lymphocyte function-associated antigen LFA-3/CD58 and BCM1/CD48 to mediate adhesion between T-cells and other cell types. CD2 is implicated in the activation of T-cells and its cytoplasmic domain is involved in the signaling transduction. The complex of CD2 and LFA-3 plays an important role in enhancing the adhesion of T cells to target cells, promoting hyperplasia, and triggering the activation of T lymphocytes.
Gene Symbol: CD2; T11; Leu-5; SRBC; LFA-2
NCBI Gene ID: 914
Uniprot Entry: P06729
Construct Details: Full length human CD2 gene is subcloned into the Lentiviral expression vector pLTC with an upstream CMV promoter and with or without a selection marker, which can be used for both transient and stable expression in mammalian cells. It can be co-transfected with the LentiPAK DNA mix (SKU# LP-001) into HEK293 cells to produce high titer lentiviral particles. Multiple choices of a stable antibiotic selection marker are available.
Vector Type: pLTC or pLTC-IRES-Marker (lentiviral expression vector containing a heterologous CMV promoter, see the vector map above)
Formulation: Supplied with 10 μg of endotoxin-free plasmid DNA at 200 ng/μl in sterile TE buffer (pH8.0)
Gene Insert Size: 1056 (bp)

Primers: Gene insert coding sequence can be confirmed by following primers:
(a) forward (or 5'-end) primer: 5’-CAAATGGGCGGTAGGCGTG-3’;
(b) reverse (or 3'-end) primer: 5’-CCAGGTTTCCGGGCCCTCAC-3’
Note: The sequence may vary due to the naturally occurring variants or mutations, such as single nucleotide polymorphism, or the artifact during PCR amplification. High fidelity PCR systems should always be used.
