Human Sox2 Full-length Gene in Lentivector, Endotoxin-free DNA
SOX2 (sex determining region Y-box 2) is a transcription factor that is essential for maintaining self-renewal, or pluripotency, of undifferentiated embryonic and neural stem cells. Sox2 is a member of the Sox family of transcription factors, which have been shown to play key roles in many stages of mammalian development. This protein family shares highly conserved DNA binding domains known as HMG (High-mobility group) box domains. Mutations in this gene have been associated with optic nerve hypoplasia and with syndromic microphthalmia, a severe form of structural eye malformation. This gene lies within an intron of another gene called SOX2 overlapping transcript (SOX2OT). SOX2 forms a trimeric complex with OCT4 on DNA and controls the expression of a number of genes involved in embryonic development. OCT4, SOX2, c-Myc and KLF4 ogether are sufficient to reprogram differentiated cells to an embryonic-like state designated iPS (induced pluripotent stem) cells. iPS cells exhibit the morphology and growth properties of ES cells and express ES cell marker genes. Sox2 holds great promise in research involving induced pluripotency, an emerging and promising field of regenerative medicine. Overexpression of SOX2 has also been implicated in multiple cancer types.

Construct Details: Full length human SOX2 gene is subcloned into the Lentiviral expression vector pLTC with an upstream CMV promoter and with or without a selection marker, which can be used for both transient and stable expression in mammalian cells. It can be co-transfected with the LentiPAK DNA mix (SKU# LP-001) into HEK293 cells to produce high titer lentiviral particles. Multiple selection markers are available.
