Description

CD142, also known as TF (tissue factor) and F3 (coagulation factor III), is a single-pass type I transmembrane glycoprotein belonging to the tissue factor family. CD142/TF contains 3 WKS motifs in its extracellular domain (ECD) in addition to a transmembrane (TM) domain and a short cytoplasmic tail.  CD142/TF plays a major role in normal hemostasis by initiating the cell-surface assembly and propagation of the coagulation protease cascade. CD142/TF acts as a high-affinity receptor for the coagulation factor VII and the [CD142:VII] complex activates factors IX or X by specific limited proteolysis. Platelets and monocytes are known to express CD142/TF under pro-coagulatory and pro-inflammatory stimuli. Unlike the other cofactors, which circulate as nonfunctional precursors, CD142/TF is a potent initiator that is fully functional when expressed on cell surfaces, e.g., on monocytes. CD142/TF also plays a role in HIV-associated coagulopathy.  In addition platelet-dependent monocyte expression of CD142/TF is associated with COVID-19 (Coronavirus Disease 2019)  severity and mortality.
