Description

TF (Transferrin), also known as Serotransferrin and Siderophilin, is a secreted glycoprotein that belongs to the transferrin family. The TF gene resides in the same region of chromosome 3 as other members of the transferrin family. TF shares sequence similarity and iron-binding properties with other family members. It contains two transferrin-like motifs and each binds a single atom of iron in association with the binding of an anion, usually bicarbonate. TF is responsible for the transport of iron from sites of absorption and heme degradation to those of storage and utilization. TF plays a further role in stimulating cell proliferation.  TF may also have a physiologic role as granulocyte/pollen-binding protein (GPBP) involved in the removal of certain organic matter and allergens from serum. The mutations in the TF gene are associated with Atransferrinemia (ATRAF), a rare autosomal recessive disorder characterized by abnormal synthesis of transferrin leading to iron overload and microcytic hypochromic anemia.

Gene Symbol: TF; Transferrin; Serotransferrin; TFQTL1; PRO1557; PRO2086; HEL-S-71p; PRO1400

NCBI Gene ID: 7018

Uniprot Entry: P02787

Construct Details: Full length human TF gene is subcloned into the Lentiviral expression vector pLTC with an upstream CMV promoter and with or without a selection marker, which can be used for both transient and stable expression in mammalian cells.  It can be co-transfected with the LentiPAK DNA mix (SKU# LP-001) into HEK293 cells to produce high titer lentiviral particles. Multiple choices of a stable antibiotic selection marker are available.

Vector Type: pLTC or pLTC-IRES-Marker (lentiviral expression vector containing a heterologous CMV promoter +/- a selection marker, see the vector map above)

Formulation: Supplied with 10 μg of endotoxin-free plasmid DNA at 200 ng/μl in sterile TE buffer (pH8.0)

Primers: Gene insert coding sequence can be confirmed by following primers:

(a) forward (or 5'-end) primer: 5’-CAAATGGGCGGTAGGCGTG-3’;

(b) reverse (or 3'-end) primer: 5’-CCAGGTTTCCGGGCCCTCAC-3’

Note: The sequence may vary due to the naturally occurring variants or mutations, such as single nucleotide polymorphism, or the artifact during PCR amplification.  High fidelity PCR systems should always be used.
