Human ALB/Albumin/HSA Gene, Full-length cDNA Clone in Lentivector, Endotoxin-free DNA
ALB (Albumin) or HSA (Human Serum Albumin) is the most abundant protein found in human blood. Human ALB gene encodes a preproprotein that is further proteolytically processed to generate the mature form. ALB regulates blood plasma colloid osmotic pressure and acts as a carrier protein for a wide range of molecules, including hormones, bilirubin, fatty acids, and metabolites, as well as exogenous drugs. ALB is a major zinc transporter in plasma, typically binds ~80% of all plasma zinc. ALB also binds ~45% of circulating calcium and magnesium in plasma. The rank order of its affinity is zinc > calcium > magnesium. ALB contains at least 2 calcium-binding sites and the shareds binding site between zinc and calcium at residue Asp-273. Additionally, ALB exhibits an esterase-like activity with broad substrate specificity. ALB binds to the bacterial siderophore enterobactin and inhibits enterobactin-mediated iron uptake of E.coli from ferric transferrin. The mutations in the ALB gene are associated with FDAH (Familial Dysalbuminemic Hyperthyroxinemia) and ANALBA (Analbuminemia) conditions.
Gene Symbol: ALB; Albumin; HSA; FDAHT; PRO0883; PRO0903; PRO1341
NCBI Gene ID: 213
Uniprot Entry: P02768
Construct Details: Full length human ALB is subcloned into the Lentiviral expression vector pLTC with an upstream CMV promoter and with or without a selection marker, which can be used for both transient and stable expression in mammalian cells. It can be co-transfected with the LentiPAK DNA mix (SKU# LP-001) into HEK293 cells to produce high titer lentiviral particles. Multiple choices of a stable antibiotic selection marker are available.
Vector Type: pLTC or pLTC-IRES-Marker (lentiviral expression vector containing a heterologous CMV promoter, see the vector map above)
Formulation: Supplied with 10 μg of endotoxin-free plasmid DNA at 200 ng/μl in sterile TE buffer (pH8.0)
Gene Insert Size: 1830 (bp)

Primers: Gene insert coding sequence can be confirmed by following primers:
(a) forward (or 5'-end) primer: 5’-CAAATGGGCGGTAGGCGTG-3’;
(b) reverse (or 3'-end) primer: 5’-CCAGGTTTCCGGGCCCTCAC-3’
Note: The sequence may vary due to the naturally occurring variants or mutations, such as single nucleotide polymorphism, or the artifact during PCR amplification. High fidelity PCR systems should always be used.
