CLTRN (collectrin) or TMEM27 (Transmembrane protein 27) is a single-pass type 1 transmembrane protein that is important for trafficking amino acid transporters to the apical brush border of proximal tubules. CLTRN binds to amino acid transporters SLC6A18 and SLC6A19 and regulates their expression on the plasma membrane. It may also play a role in trafficking amino acid transporters SLC3A1 and SLC7A9 to the renal cortical cell membrane.  CLTRN may control insulin exocytosis by regulating formation of the SNARE (soluble N-ethylmaleimide-sensitive-factor attachment protein receptor) complex in pancreatic beta cells. The extracellular domain of CLTRN may be shed from the membrane in pancreatic beta cells by BACE2 (beta-secretase 2). This leads to the release of soluble CLTRN and a corresponding cell-associated C-terminal fragment that is later cleaved by gamma-secretase.  CLTRN may function as a stimulator of beta cell replication and the shedding process may inactivate CLTRN.

Construct Details: Full length human CLTRN gene is subcloned into the Lentiviral expression vector pLTC with an upstream CMV promoter and with or without a selection marker, which can be used for both transient and stable expression in mammalian cells.  It can be co-transfected with the LentiPAK DNA mix (SKU# LP-001) into HEK293 cells to produce high titer lentiviral particles. Multiple choices of a stable antibiotic selection marker are available.

The product is shipped at 4°C. Upon receipt, centrifuge the product briefly before opening the vial. It is recommended to store small aliquots at the temperature below –20°C for long-term storage and the product is stable for 6 months.
