Description

Angiotensin-converting enzymes including ACE and ACE2 are a central component of the renin-angiotensin system (RAS) that controls blood pressure by regulating the volume of fluids in the body. Both ACE & ACE2 are single-pass type I membrane proteins that belong to the peptidase M2 family.  Unlike ACE, the extracellular region of ACE2 contains only 1 zinc-binding metallopeptidase domain (19-611aa) and a C-terminal domain (612-740aa) that is 48% identical to human collectrin.  Its cytoplasmic tail contains several motifs such as LIR, PDZ-binding, PTB and endocytic sorting signals allowing interactions with proteins that mediate endocytic trafficking and autophagy.  The secreted ACE2 catalyzes the cleavage of angiotensin I into angiotensin 1-9, and angiotensin II into the vasodilator angiotensin 1-7. ACE2 is expressed in various human organs and may play a role in the regulation of cardiovascular and renal function, as well as fertility. In addition, ACE2 is a functional receptor for the spike glycoprotein of the human coronavirus HCoV-NL63 and the human severe acute respiratory syndrome coronaviruses, SARS-CoV and SARS-CoV-2, the latter is the causative agent of coronavirus disease-2019 (COVID-19).

Construct Details: Full length human ACE2 gene is subcloned into the Lentiviral expression vector pLTC with an upstream CMV promoter and with or without a selection marker, which can be used for both transient and stable expression in mammalian cells.  It can be co-transfected with the LentiPAK DNA mix (SKU# LP-001) into HEK293 cells to produce high titer lentiviral particles. Multiple choices of a stable antibiotic selection marker are available.

The product is shipped at 4°C. Upon receipt, centrifuge the product briefly before opening the vial. It is recommended to store small aliquots at the temperature below –20°C for long-term storage and the product is stable for 6 months. 

 

 

Use a manual defrost freezer and avoid repeated freeze-thaw cycles.
