Description

Killer cell immunoglobulin-like receptors (KIRs), also known as Killer-cell Inhibitory Receptors, are single pass type I transmembrane glycoproteins that belong to the immunoglobulin superfamily (IgSF).  The KIR genes are highly homologous and polymorphic.  Most of them are found in a cluster on chromosome 19q13.4 within the leukocyte receptor complex (LRC). KIRs are expressed on natural killer (NK) cells and subsets of T cells. KIR proteins are composed of 2 or 3 extracellular Ig domains, a transmembrane region and cytoplasmic tail which can in turn be short or long. The KIR proteins are thus classified by the number of Ig-domains (2D or 3D) and by whether they have a long (L) or short (S) cytoplasmic domain. KIR proteins with the long cytoplasmic domain transduce inhibitory signals upon ligand binding via an immune tyrosine-based inhibitory motif (ITIM).  In contrast KIR proteins with the short cytoplasmic domain lack the ITIM motif and instead transduce activating signals. The ligands for several KIR proteins are subsets of HLA class I molecules; thus, KIR proteins are thought to play an important role in regulation of the immune response. KIR2DL subgroup proteins, including KIR2DL1-3 are receptors on NK cells for HLA-C alleles and inhibit the activity of NK cells thus preventing cell lysis. KIR2DL4 is a receptor for non-classical major histocompatibility class Ib HLA-G and triggers NK cell senescence-associated secretory phenotype. KIR2DL4 may play a role in balancing tolerance and antiviral-immunity at maternal-fetal interface by keeping in check the effector functions of NK, CD8+ T cells and B cells.

NCBI Gene ID: 3805

Uniprot Entry: Q99706

Construct Details: Full length human KIR2DL4 gene (encoding Uniprot# Q99706-3) is subcloned into the Lentiviral expression vector pLTC with an upstream CMV promoter, which can be used for both transient and stable expression in mammalian cells. It can be co-transfected with the LentiPAK DNA mix (SKU# LP-001) into HEK293 cells to produce high titer lentiviral particles.

Vector Type: pLTC (lentiviral expression vector containing a heterologous CMV promoter, see the vector map above)

Formulation: Supplied with 10 μg of endotoxin-free plasmid DNA at 200 ng/μl in sterile TE buffer (pH8.0)

Primers: Gene insert coding sequence can be confirmed by following primers:

(a) forward (or 5'-end) primer: 5’-CAAATGGGCGGTAGGCGTG-3’;

(b) reverse (or 3'-end) primer: 5’-CGGGAAGCAATAGCATGATA-3’

Note: The sequence may vary due to the naturally occurring variants or mutations, such as single nucleotide polymorphism, or the artifact during PCR amplification.  High fidelity PCR systems should always be used.
