Description

Telomerase is a ribonucleoprotein polymerase essential for the replication of chromosome termini in most eukaryotes. TERT (Telomerase Reverse Transcriptase) represents the catalytic component of Telomerase holoenzyme complex.  TERT acts as a reverse transcriptase that adds simple sequence repeats “TTAGGG” to chromosome ends by copying a template sequence within the RNA component of the holoenzyme.  Telomerase activity is regulated by many factors including telomerase complex-associated proteins, chaperones and polypeptide modifiers. Telomerase expression plays an important role in cellular senescence, as it is repressed in postnatal somatic cells leading to progressive shortening of telomeres. Activation of telomerase has been implicated in cell immortalization and oncogenesis.  It also plays important roles in aging and antiapoptosis.  Genetic variations in the TERT gene are associated with a number of diseases including aplastic anemia, coronary artery disease, dyskeratosis congenita, pulmonary fibrosis, and melanoma.

Construct Details: Full length human TERT gene is subcloned into the Lentiviral expression vector pLTC with an upstream CMV promoter and with or without an antibiotic selection marker, which can be used for both transient and stable expression in mammalian cells.  It can be co-transfected with the LentiPAK DNA mix (SKU# LP-001) into HEK293 cells to produce high titer lentiviral particles. Multiple choices of a stable antibiotic selection marker are available.

The product is shipped at 4°C. Upon receipt, centrifuge the product briefly before opening the vial. It is recommended to store small aliquots at the temperature below –20°C for long-term storage and the product is stable for 6 months.
