Human CCRL2/CCR11/CKRX/HCR Gene, Full-length cDNA in Lentivector, Endotoxin-free DNA
CCRL2 (C-C chemokine receptor-like 2) is a 7-transmembrane protein of the G protein-coupled receptor 1 (GPCR1) family and most closely related to CCR1. CCRL2 acts as a receptor for C-C type chemokines. Chemokines and their receptors mediated signal transduction are critical for the recruitment of effector immune cells to the site of inflammation. The CCRL2 gene and other chemokine receptor genes, including CCR1, CCR2, CCR3, CCR5 and CCXCR1, are found to form a gene cluster on chromosome 3p. CCRL2 is expressed at high levels in primary neutrophils and primary monocytes, and is further upregulated on neutrophil activation and during monocyte to macrophage differentiation. The function of the CCRL2 gene is unknown. It may act as a receptor for CCL19 and chemerin/RARRES2. However CCRL2 does not appear to be a signaling receptor. It may have a role in modulating chemokine-triggered immune responses by capturing and internalizing CCL19 or by presenting RARRES2 ligand to CMKLR1, a functional signaling receptors. CCRL2 may play a critical role in the development of Th2 responses.
Gene Symbol: CCRL2; CCR11; HCR; CKRX; CRAM; ACKR5; CRAM-A; CRAM-B
NCBI Gene ID: 9034
Uniprot Entry: O00421
Construct Details: Full length human CCRL2/CCR11 gene is subcloned into the Lentiviral expression vector pLTC with an upstream CMV promoter and with or without an antibiotic selection marker, which can be used for both transient and stable expression in mammalian cells. It can be co-transfected with the LentiPAK DNA mix (SKU# LP-001) into HEK293 cells to produce high titer lentiviral particles. Multiple choices of a stable antibiotic selection marker are available.
Vector Type: pLTC or pLTC-IRES-Marker (lentiviral expression vector containing a heterologous CMV promoter +/- a selection marker, see the vector map above)
Formulation: Supplied with 10 μg of endotoxin-free plasmid DNA at 200 ng/μl in sterile TE buffer (pH8.0)
Gene Insert Size: 1035 (bp)

Primers: Gene insert coding sequence can be confirmed by following primers:
(a) forward (or 5'-end) primer: 5’-CAAATGGGCGGTAGGCGTG-3’;
(b) reverse (or 3'-end) primer: 5’-CCAGGTTTCCGGGCCCTCAC-3’
Note: The sequence may vary due to the naturally occurring variants or mutations, such as single nucleotide polymorphism, or the artifact during PCR amplification. High fidelity PCR systems should always be used.
