Description

CCR4 (C-C chemokine receptor type 4) is a 7-transmembrane protein of the G protein-coupled receptor 1 (GPCR1) family. CCR4 acts as a receptor for C-C type chemokines. Chemokines and their receptors mediated signal transduction are critical for the recruitment of effector immune cells to the site of inflammation.  CCR4 is a high affinity receptor for the C-C type chemokines CCL17/TARC, CCL22/MDC and CKLF isoform 1/CKLF1. CCR4 transduces the signal through a phosphatidylinositol-calcium second messenger system. CCR4 may function as a chemoattractant homing receptor on circulating memory lymphocytes and as a coreceptor for some primary HIV-2 isolates. In the CNS, CCR4 may mediate hippocampal-neuron survival.

Gene Symbol: CCR4; CD194; CKR4; CMKBR4; K5-5; CMKBR4; ChemR13; CC-CKR-4; HGCN:14099

NCBI Gene ID: 1233

Construct Details: Full length human CCR4 gene is subcloned into the Lentiviral expression vector pLTC with an upstream CMV promoter and with or without an antibiotic selection marker, which can be used for both transient and stable expression in mammalian cells.  It can be co-transfected with the LentiPAK DNA mix (SKU# LP-001) into HEK293 cells to produce high titer lentiviral particles. Multiple choices of a stable antibiotic selection marker are available.

Vector Type: pLTC or pLTC-IRES-Marker (lentiviral expression vector containing a heterologous CMV promoter +/- a selection marker, see the vector map above)

Formulation: Supplied with 10 μg of endotoxin-free plasmid DNA at 200 ng/μl in sterile TE buffer (pH8.0)

Primers: Gene insert coding sequence can be confirmed by following primers:

(a) forward (or 5'-end) primer: 5’-CAAATGGGCGGTAGGCGTG-3’;

(b) reverse (or 3'-end) primer: 5’-CCAGGTTTCCGGGCCCTCAC-3’

Note: The sequence may vary due to the naturally occurring variants or mutations, such as single nucleotide polymorphism, or the artifact during PCR amplification.  High fidelity PCR systems should always be used.
