Human CEACAM4/CGM7/NCA Gene, Full-length cDNA Clone in Lentivector, Endotoxin-free DNA
CEACAM4 (carcinoembryonic antigen (CEA)-related cell adhesion molecule 4) and Carcinoembryonic antigen CGM7, is a single pass type I transmembrane protein of the CEA family of the immunoglobulin superfamily (IgSF). CEA family members are either transmembrane or GPI-anchored within two subgroups: the CEACAMs and the pregnancy-specific glycoproteins that are used by several bacterial pathogens to bind and invade host cells. Members of the CEA family consist of a single N domain, with structural similarity to the Ig variable (IgV) domains, followed by a variable number of Ig constant-like 1 (IgC1) and/or 2 (IgC2) domains. In human, 7 CEACAMs have been identified including 3 transmembrane proteins (CEACAM1, 3, 4) and 4 GPI-anchored molecules (CEACAM5-8). Human CEACAM4 contains 1 Ig-like V-type (IgV) motif in the N-terminal 120 aa extracellular domain (ECD), followed by a transmembrane domain (TM) and a 68 aa cytoplasmic tail with the ITAM-like sequence. The ITAM becomes tyrosine phosphorylated by SRC family PTKs upon ligand-mediated receptor clustering and allows to initiate phagocytosis of bound ligand. CEACAM4 is a Granulocyte orphan receptor that acts as an trigger efficient phagocytosis of attached particles.
Gene Symbol: CEACAM4; NCA; CGM7; W236
NCBI Gene ID: 1089
Uniprot Entry: O75871
Construct Details: Full length human CEACAM4 gene is subcloned into the Lentiviral expression vector pLTC with an upstream CMV promoter and with or without a selection marker, which can be used for both transient and stable expression in mammalian cells. It can be co-transfected with the LentiPAK DNA mix (SKU# LP-001) into HEK293 cells to produce high titer lentiviral particles. Multiple choices of a stable antibiotic selection marker are available.
Vector Type: pLTC or pLTC-IRES-Marker (lentiviral expression vector containing a heterologous CMV promoter, see the vector map above)
Formulation: Supplied with 10 μg of endotoxin-free plasmid DNA at 200 ng/μl in sterile TE buffer (pH8.0)
Gene Insert Size: 735 (bp)

Primers: Gene insert coding sequence can be confirmed by following primers:
(a) forward (or 5'-end) primer: 5’-CAAATGGGCGGTAGGCGTG-3’;
(b) reverse (or 3'-end) primer: 5’-CCAGGTTTCCGGGCCCTCAC-3’
Note: The sequence may vary due to the naturally occurring variants or mutations, such as single nucleotide polymorphism, or the artifact during PCR amplification. High fidelity PCR systems should always be used.
