Description

C7orf15 (chromosome 7 open reading frame 15) also know as PVRIG (Poliovirus receptor-related immunoglobulin domain-containing protein) and CD112R may encode a 3-pass transmembrane protein. It was named PVRIG for the homology observed between its second exon and the variable immunoglobulin (Ig) domain of the polio virus receptor (PVR/CD155) and polio virus receptor-like (PVRL) genes.  C7orf15 is expressed in some types of immune cells. It is expressed at low levels on the surface of freshly isolated T-cells and natural killer (NK) cells but not detected in B-cells, naive or helper T-cells, monocytes, nor neutrophils. Its expression in T cells is up-regulated by activation.  C7orf15 is the cell surface receptor for NECTIN2 and competes with CD226 for NECTIN2-binding.  It may act as a coinhibitory receptor that suppresses T-cell receptor-mediated signals. Following interaction with NECTIN2, C7orf15 inhibits T-cell proliferation.

Construct Details: Full length human PVRIG gene is subcloned into the Lentiviral expression vector pLTC with an upstream CMV promoter and with or without an antibiotic selection marker, which can be used for both transient and stable expression in mammalian cells.  It can be co-transfected with the LentiPAK DNA mix (SKU# LP-001) into HEK293 cells to produce high titer lentiviral particles. Multiple choices of a stable antibiotic selection marker are available.

The product is shipped at 4°C. Upon receipt, centrifuge the product briefly before opening the vial. It is recommended to store small aliquots at the temperature below –20°C for long-term storage and the product is stable for 6 months.
