Human ATP4B Gene, Full-length cDNA Clone in Lentivector, Endotoxin-free DNA
ATP4B (ATPase H+/K+ transporting beta subunit) is a single-pass type II transmembrane glycoprotein beloning to the family of Na+/K+ and H+/K+ ATPases beta chain proteins. Na+/K+ -ATPase is an integral membrane protein responsible for establishing and maintaining the electrochemical gradients of Na and K ions across the plasma membrane. These gradients are essential for osmoregulation, for sodium-coupled transport of a variety of organic and inorganic molecules, and for electrical excitability of nerve and muscle. This ATPase is composed of three subunits: a large catalytic subunit alpha (catalytic), a smaller glycoprotein subunit beta and gamma. The beta subunit regulates, through assembly of alpha/beta heterodimers, the number of sodium pumps transported to the plasma membrane. ATP4B is the beta subunit of the gastric H+, K+-ATPase. The gastric H+, K+-ATPase is a proton pump that catalyzes the hydrolysis of ATP coupled with the exchange of H(+) and K(+) ions across the plasma membrane. It is also responsible for gastric acid secretion. ATP4B may be required for stabilization and maturation of the catalytic proton pump alpha subunit. The C-terminal lobe of ATP4B folds into an immunoglobulin-like domain and may mediate cell adhesion and establish epithelial cell polarity.

Construct Details: Full length human ATP4B is subcloned into the Lentiviral expression vector pLTC with an upstream CMV promoter and with or without an antibiotic selection marker, which can be used for both transient and stable expression in mammalian cells. It can be co-transfected with the LentiPAK DNA mix (SKU# LP-001) into HEK293 cells to produce high titer lentiviral particles. Multiple choices of a stable antibiotic selection marker are available.
