Description

ATP1B1 (ATPase Na+/K+ transporting subunit beta 1) is a single-pass type II transmembrane glycoprotein beloning to the family of Na+/K+ and H+/K+ ATPases beta chain proteins. Na+/K+ -ATPase is an integral membrane protein responsible for establishing and maintaining the electrochemical gradients of Na and K ions across the plasma membrane. These gradients are essential for osmoregulation, for sodium-coupled transport of a variety of organic and inorganic molecules, and for electrical excitability of nerve and muscle. This ATPase is composed of three subunits: a large catalytic subunit alpha (catalytic), a smaller glycoprotein subunit beta and gamma. The beta subunit regulates, through assembly of alpha/beta heterodimers, the number of sodium pumps transported to the plasma membrane. ATP1B1 is a non-catalytic component of the active enzyme, which catalyzes the hydrolysis of ATP coupled with the exchange of Na+ and K+ ions across the plasma membrane. The exact function of the ATP1B1 subunit is not known and the C-terminal lobe of ATP1B1 folds into an immunoglobulin-like domain and may mediate cell adhesion and establish epithelial cell polarity.

Construct Details: Full length human ATP1B1 is subcloned into the Lentiviral expression vector pLTC with an upstream CMV promoter and with or without an antibiotic selection marker, which can be used for both transient and stable expression in mammalian cells.  It can be co-transfected with the LentiPAK DNA mix (SKU# LP-001) into HEK293 cells to produce high titer lentiviral particles. Multiple choices of a stable antibiotic selection marker are available.

The product is shipped at 4°C. Upon receipt, centrifuge the product briefly before opening the vial. It is recommended to store small aliquots at the temperature below –20°C for long-term storage and the product is stable for 6 months.
