Description

BSG or CD147 is a 385 amino acid (aa) single pass type I transmembrane protein of the Basigin family of the Immunoglobulin superfamily (IgSF).  BSG contains an N-terminal signal peptide, a 186-aa extracellular domain (ECD) with 1 Ig-like C2-type and 1 Ig-like V-type domains and 3 potential N-linked glycosylation sites, a transmembrane domain (TM), and a 41-aa cytoplasmic tail. BSG forms homooligomers in a cis-dependent manner on the plasma membrane.  BSG interacts with ATP1B2, MAG and L1CAM.  BSG is important in spermatogenesis, embryo implantation, neural network formation, and tumor progression. BSG plays an important role in targeting the monocarboxylate transporters SLC16A1, SLC16A3 and SLC16A8 to the plasma membrane. BSG stimulates adjacent fibroblasts to produce matrix metalloproteinases (MMPS). BSG is up-regulated in gliomas and its expression levels correlate with malignant potential of the tumor.

Gene Symbol: BSG; CD147; OK; 5F7; TCSF; BASIGIN; EMMPRIN

NCBI Gene ID: 682

Uniprot Entry: P35613

Construct Details: Full length human BSG/CD147 gene is subcloned into the Lentiviral expression vector pLTC with an upstream CMV promoter and with or without an antibiotics selection marker, which can be used for both transient and stable expression in mammalian cells.  It can be co-transfected with the LentiPAK DNA mix (SKU# LP-001) into HEK293 cells to produce high titer lentiviral particles.

Vector Type: pLTC or pLTC-Marker (see the vector map above)

Formulation: Supplied with 10 μg of endotoxin-free plasmid DNA at 200 ng/μl in sterile TE buffer (pH8.0)

Primers: Gene insert coding sequence can be confirmed by following primers:

(a) forward (or 5'-end) primer: 5’-CAAATGGGCGGTAGGCGTG-3’;

(b) reverse (or 3'-end) primer: 5’-CCAGGTTTCCGGGCCCTCAC-3’

Note: The sequence may vary due to the naturally occurring variants or mutations, such as single nucleotide polymorphism, or the artifact during PCR amplification.  High fidelity PCR systems should always be used.
