Human ABCG5/STSL Gene, Full-length cDNA Clone in Lentivector, Endotoxin-free DNA
ABCG5 (ATP-binding cassette sub-family G member 5) is a six-pass type II transmembrane glycoprotein beloning to the ABC transporter superfamily. ABC proteins transport various molecules across extra- and intra-cellular membranes. ABC genes are divided into seven distinct subfamilies (ABC1, MDR/TAP, MRP, ALD, OABP, GCN20, White/ABCG). ABCG4 is a member of the White/ABCG subfamily. ABCG1 plays an important role in cellular cholesterol homeostasis. ABCG5 functions as a half-transporter to limit intestinal absorption and promote biliary excretion of sterols. ABCG5 is expressed in a tissue-specific manner in the liver, colon, and intestine. Mutations in the ABCG5 gene may contribute to sterol accumulation and atheroschlerosis, and have been observed in patients with sitosterolemia (STSL), an autosomal recessive disorder characterized by increased intestinal absorption of all sterols including cholesterol, plant and shellfish sterols, and decreased biliary excretion of dietary sterols into bile. ABCG5 may form heterodimers with ABCG8 or be tightly coupled to ABCG8 along a pathway regulating diatery-sterol absorption and excretion.
Gene Symbol: ABCG5; STSL
NCBI Gene ID: 64240
Uniprot Entry: Q9H222
Construct Details: Full length human ABCG5 is subcloned into the Lentiviral expression vector pLTC with an upstream CMV promoter and with or without a selection marker, which can be used for both transient and stable expression in mammalian cells. It can be co-transfected with the LentiPAK DNA mix (SKU# LP-001) into HEK293 cells to produce high titer lentiviral particles. Multiple choices of a stable antibiotic selection marker are available.
Vector Type: pLTC or pLTC-IRES-Marker (lentiviral expression vector containing a heterologous CMV promoter +/- a selection marker, see the vector map above)
Formulation: Supplied with 10 μg of endotoxin-free plasmid DNA at 200 ng/μl in sterile TE buffer (pH8.0)
Gene Insert Size: 1956 (bp)

Primers: Gene insert coding sequence can be confirmed by following primers:
(a) forward (or 5'-end) primer: 5’-CAAATGGGCGGTAGGCGTG-3’;
(b) reverse (or 3'-end) primer: 5’-CCAGGTTTCCGGGCCCTCAC-3’
Note: The sequence may vary due to the naturally occurring variants or mutations, such as single nucleotide polymorphism, or the artifact during PCR amplification. High fidelity PCR systems should always be used.
