Human CD320/8D6A/TCBLR Gene, Full-length cDNA in Lentivector, Endotoxin-free DNA
CD320 is a 282 amino acid (aa) single pass type I transmembrane glycoprotein that acts as a receptor for transcobalamin saturated with cobalamin (TCbl). CD320 contains an N-terminal signal peptide, a 194-aa extracellular domain (ECD) with 2 LDL-receptor class A domains and 3 potential glycosylation sites, a transmembrane domain (TM), and a 32-aa short cytoplasmic tail. CD320 mediates the cellular uptake of transcobalamin bound cobalamin (vitamin B12), and is involved in B-cell proliferation and immunoglobulin secretion. CD320 is detected in the germinal center (GC) of lymphoid follicles. CD320 is expressed abundantly on follicular dendritic cells (FDC) and mediates interaction with germinal center B cells CD320 functions as a c-ostimulator to promote B cell responses to antigenic stimuli and promotes B cell differentiation and proliferation as well as germinal center-B (GC-B) cells differentiate into memory B-cells and plasma cells (PC) through interaction with T-cells and follicular dendritic cells (FDC). CD320 augments the proliferation of PC precursors generated by IL-10. Mutations in the CD320 gene are associated with methylmalonic aciduria, a metabolic disorder characterized by increased blood C3-acylcarnitine levels, elevated methylmalonate and homocysteine, and low uptake of transcobalamin-bound cobalamin.

Construct Details: Full length human CD320 gene is subcloned into the Lentiviral expression vector pLTC with an upstream CMV promoter and with or without an antibiotic selection marker, which can be used for both transient and stable expression in mammalian cells. It can be co-transfected with the LentiPAK DNA mix (SKU# LP-001) into HEK293 cells to produce high titer lentiviral particles. Multiple choices of a stable antibiotic selection marker are available.
