Human SIGLEC5/CD170/CD33L2/OBBP2 Gene, Full-length cDNA in Lentivector, Endotoxin-free DNA
SIGLEC5 or CD170 is a single-pass type I transmembrane protein belonging to the sialic acid binding Ig-like lectin (SIGLEC) family in the immunoglobulin superfamily (IgSF). SIGLECs are characterized by an N-terminal Ig-like V-type (IgV)domain that mediates sialic acid binding, followed by varying numbers (2 to 17) of Ig-like C2-type (IgC2) domains in the extracellular region, a transmembrane region, and a cytoplasmic tail with intracellular immunoreceptor tyrosine-based inhibitory motifs (ITIM). They can be classified into two subgroups: Siglecs-1, -2, and -4 subgroup, and a Siglec-3/CD33-related subgroup (Siglecs-3, and -5 through -13), defined by sequence similarity and clustered gene localization. SIGLECs are widely expressed on hematopoietic cells, often in a cell-type-specific manner. SIGLEC5 contains only 2 IgC2 domains and inhibits the activation of several cell types including monocytes, macrophages and neutrophils. SIGLEC5 is a putative adhesion molecule that mediates sialic-acid dependent binding to cells. It binds equally to alpha-2,3-linked and alpha-2,6-linked sialic acid. Binding of group B Streptococcus (GBS) to the encoded protein plays a role in GBS immune evasion.

Construct Details: Full length human SIGLEC5/CD170 gene is subcloned into the Lentiviral expression vector pLTC with an upstream CMV promoter and with or without an antibiotics selection marker, which can be used for both transient and stable expression in mammalian cells. It can be co-transfected with the LentiPAK DNA mix (SKU# LP-001) into HEK293 cells to produce high titer lentiviral particles.

The product is shipped at 4°C. Upon receipt, centrifuge the product briefly before opening the vial. It is recommended to store small aliquots at the temperature below –20°C for long-term storage and the product is stable for 6 months.
