Control RFP Reporter Gene in Lentivector, EF1a-promoter, Endotoxin-free DNA
RFP (red fluorescent protein) is a monomeric fluorophore with peak absorption and emission at 587 nm and 610 nm, respectively. RFP derives from a protein isolated from Discosoma sp., while other fluorescent proteins in the green range are often variants of GFP from Aequorea victoria. Like other fluorescent proteins, RFP contains a chromophore that is formed by autocatalytic backbone condensation between Xaa-N and Gly-N+2, oxidation of Tyr-N+1 to didehydrotyrosine, and formation of a double bond to the alpha-amino nitrogen of residue Xaa-N. Maturation of the chromophore requires nothing other than molecular oxygen. RFP is resistant to photobleaching and is stable. It matures quickly, allowing it to be visualised soon after translation. RFP becomes a useful tool for making chimeric proteins, where it can be fused to a broad variety of proteins at either the N- or C-terminus. They have been expressed in most known cell types and are used as a noninvasive fluorescent marker in living cells and organisms. They enable a wide range of applications where they have functioned, for example, as a cell lineage tracer, reporter of gene expression, or as a measure of protein-protein interactions. The pre-packaged lentiviral particles can be transduced into any mammalian cell type for constitutively expressing RFP. Multiple choices of antibiotic stable-selection markers are available.

Construct Details: RFP is cloned into the Lentiviral expression vector pLTC with an EF1a promoter and with or without a selection marker, which can be used for both transient and stable expression in mammalian cells. It was co-transfected with the LentiPAK DNA mix (SKU# LP-001) into HEK293 cells to produce high titer lentiviral particles.

Note: The sequence may vary due to the naturally occurring variants or mutations, such as single nucleotide polymorphism, or the artifact during PCR amplification. High fidelity PCR systems should always be used.

The product is shipped at 4°C. Upon receipt, centrifuge the product briefly before opening the vial. It is recommended to store small aliquots at the temperature below –20°C for long-term storage and the product is stable for 6 months.
