Human CDH2/CD325/NCAD/Cadherin2 Gene, Full-length cDNA in Lentivector, Endotoxin-free DNA
Cadherins are calcium-dependent cell adhesion proteins that preferentially interact with themselves in a homophilic manner in connecting cells. Cadherins thus contribute to the sorting of heterogeneous cell types. Cadherin-2 or CDH2 is a 906 amino acid (aa) single pass type I transmembrane protein of the cadherin superfamily. CDH2 contains an N-terminal signal peptide, a 565-aa extracellular domain (ECD) with 5 cadherin domains and 7 potential glycosylation sites, a transmembrane domain (TM), and a 161-aa highly conserved cytoplasmic tail. CDH2 is identified in a complex containing FGFR4, NCAM1, CDH2, PLCG1, FRS2, SRC, SHC1, GAP43 and CTTN. It also interacts with CDCP1 and PCDH8. CDH2 plays a role in the establishment of left-right asymmetry, development of the nervous system and the formation of cartilage and bone. CDH2 may be leaved by MMP24, leading to the generation of a soluble 90 kDa form of ECD and a 45 kDa membrane-bound carboxy-terminal fragment 1 (CTF1), which is further cleaved by gamma-secretase into a 35 kDa. Cleavage in neural stem cells by MMP24 affects CDH2-mediated anchorage of neural stem cells, leading to modulate neural stem cell quiescence.

Construct Details: Full length human CDH2/CD325 gene is subcloned into the Lentiviral expression vector pLTC with an upstream CMV promoter and with or without a selection marker, which can be used for both transient and stable expression in mammalian cells. It can be co-transfected with the LentiPAK DNA mix (SKU# LP-001) into HEK293 cells to produce high titer lentiviral particles. Multiple choices of a stable antibiotic selection marker are available.
