Anti-CD33 Monoclonal Antibody, Human IgG1, Biotinylated, Recombinant
Myeloid cell surface antigen CD33, also known as GP67 and SIGLEC-3, is a single-pass type I transmembrane protein belonging to the sialic acid binding Ig-like lectin (SIGLEC) family in the immunoglobulin superfamily (IgSF). SIGLECs are characterized by an N-terminal Ig-like V-type domain that mediates sialic acid binding, followed by varying numbers (2 to 17) of Ig-like C2-type domains, a transmembrane region, and a cytoplasmic tail with intracellular immunoreceptor tyrosine-based inhibitory motifs (ITIM). They can be classified into two subgroups: SIGLEC-1, -2, and -4 subgroup, and a SIGLEC-3/CD33-related subgroup (SIGLEC-3, and -5 through -13), defined by sequence similarity and clustered gene localization. SIGLECs are widely expressed on hematopoietic cells, often in a cell-type-specific manner. CD33/SIGLEC-3 contains 1 Ig-like C2-type and 1 Ig-like V-type domains. CD33/SIGLEC-3 expression is restricted to cells of monocytic/myeloid lineage. It is a putative adhesion molecule that preferentially binds sialic acid with α2,6 linkage. CD33/SIGLEC-3 may act as an inhibitory receptor upon ligand induced tyrosine phosphorylation by recruiting cytoplasmic phosphatases. When cocrosslinking with FcγR1, CD33/SIGLEC-3 inhibits tyrosine phosphorylation and calcium mobilization. CD33/SIGLEC-3 can also induce apoptosis in acute myeloid leukemia (AML) cells.

Construct Details: Expressed as the combination of a heavy chain (HC) containing VH from anti-human CD33 mAb and CH region of human IgG1 and a human kappa light chain (LC) encoding VL from anti-CD33 mAb. Exists as a disulfide linked dimer of the HC and LC hetero-dimer under non-reducing condition.
Source: Mammalian cells stably expressing anti-CD33 heavy chain and light chain (in human IgG1 format) and growing in chemical-defined media with no animal components.

Formulation: Supplied at 1.0 mg/ml in sterile PBS pH7.4 without any carrier protein or preservative (the concentration determined by UV absorption with an A280nm of 1.0 for 0.73 mg/ml of IgG and verified by SDS-PAGE and Coomassie blue staining). The purified recombinant monoclonal antibody was labeled with Biotin (3-5 Biotin per IgG) using the standard procedure.

Biological Activity: Binds CD33/SIGLEC-3 (SKU#FCL0133) with high affinity (KD < 10 nM) by a functional ELISA. Recognizes cell surface human CD33 by flow cytometry, IHC staining and fluorescent microscopy. Blocks CD33 Ligand-binding and signaling activity.

The product is shipped at 4°C. Upon receipt, centrifuge the product briefly before opening the vial. It is recommended to store small aliquots at the temperature below –20°C for long-term storage and the product is stable for 3 months. The undiluted protein can be stored at 4°C for no more than 2 weeks.
Use a manual defrost freezer and avoid repeated freeze-thaw cycles.
