Human KLRK1/CD314/NKG2D Gene, Full-length cDNA in Lentivector, Endotoxin-free DNA
KLRK1 belongs to the killer cell lectin-like receptor (KLR) or NKG2 family of the C-type lectin superfamily, which is a group of single pass type II transmembrane proteins preferentially expressed in natural killer (NK) cells. NK cells are lymphocytes that can mediate lysis of certain tumor cells and virus-infected cells without previous activation. NK cells regulate specific humoral and cell-mediated immunity. The KLR/NKG2 family is characterized by the presence of a C-type lectin domain in the extracellular region at the C-terminus. KLRK1 forms a disulfide-bond linked homodimer. KLRK1 function as an activating and costimulatory receptor involved in immunosurveillance upon binding to various cellular stress-inducible ligands displayed at the surface of autologous tumor cells and virus-infected cells. KLRK1 binds to ligands belonging to various subfamilies of MHC class I-related glycoproteins including MICA, MICB, RAET1E, RAET1G, ULBP1, ULBP2, ULBP3 (ULBP2>ULBP1>ULBP3) and ULBP4. KLRK1 provides both stimulatory and costimulatory innate immune responses on activated NK cells, leading to cytotoxic activity. KLRK1 also acts as a costimulatory receptor for T-cell receptor (TCR) in CD8+ T-cell-mediated adaptive immune responses by amplifying T-cell activation.

Construct Details: Full length human KLRK1 gene is subcloned into the Lentiviral expression vector pLTC with an upstream CMV promoter and with or without a selection marker, which can be used for both transient and stable expression in mammalian cells. It can be co-transfected with the LentiPAK DNA mix (SKU# LP-001) into HEK293 cells to produce high titer lentiviral particles. Multiple choices of a stable antibiotic selection marker are available.
