Description

KLRG1 belongs to the killer cell lectin-like receptor (KLR) or NKG2 family of the C-type lectin superfamily, which is a group of single pass type II transmembrane proteins preferentially expressed in natural killer (NK) cells.  NK cells are lymphocytes that can mediate lysis of certain tumor cells and virus-infected cells without previous activation. NK cells regulate specific humoral and cell-mediated immunity. The KLR/NKG2 family is characterized by the presence of a C-type lectin domain in the extracellular region at the C-terminus. KLRG1 forms a disulfide-bond linked  homodimer. KLRG1 is expressed specifically on natural killer (NK) cells and T-cells, mainly CD8 T-cell. KLRG1 contains 1 copy of a cytoplasmic motif referred to as the immunoreceptor tyrosine-based inhibitor motif (ITIM) involved in modulation of cellular responses. KLRG1 plays an inhibitory role on natural killer (NK) cells and T-cell functions upon binding to their non-MHC ligands. KLRG1 may mediate missing self recognition by binding to a highly conserved site on classical cadherins, enabling it to monitor expression of E-cadherin/CDH1, N-cadherin/CDH2 and R-cadherin/CDH4 on target cells.

Construct Details: Full length human KLRG1 gene is subcloned into the Lentiviral expression vector pLTC with an upstream CMV promoter and with or without a selection marker, which can be used for both transient and stable expression in mammalian cells.  It can be co-transfected with the LentiPAK DNA mix (SKU# LP-001) into HEK293 cells to produce high titer lentiviral particles. Multiple choices of a stable antibiotic selection marker are available.

 

Vector Type: pLTC or pLTC-IRES-Marker (lentiviral expression vector containing a heterologous CMV promoter +/- a selection marker, see the vector map above)

 

Formulation: Supplied with 10 μg of endotoxin-free plasmid DNA at 200 ng/μl in sterile TE buffer (pH8.0)

The product is shipped at 4°C. Upon receipt, centrifuge the product briefly before opening the vial. It is recommended to store small aliquots at the temperature below –20°C for long-term storage and the product is stable for 6 months.
