Description

LILRB3 (Leukocyte immunoglobulin like receptor B3), also known as CD85a, LIR3 (Leukocyte immunoglobulin-like receptor 3), and 1ILT5 (Ig-like transcript 5), is a member of the leukocyte immunoglobulin (Ig)-like receptor (LIR) family of the Ig superfamily (IgSF).  It belongs to the subfamily B class of LIR receptors (LILRBs), which are single-pass type I trnasmembrane proteins with 2 or 4 extracellular Ig-like domains, a transmembrane domain (TM), and 2-4 cytoplasmic immunoreceptor tyrosine-based inhibitory motifs (ITIMs). LILRB3 has 4 Ig-like C2 type domains and 3 TIIMs. The phosphorylated ITIM motif can bind the SH2 domain of several SH2-containing phosphatases to modulate cellular responses.  LILRB3 is expressed predominantly on B-cells and monocytes. Like LILRB1 & LILRB, LILRB3 binds to MHC class I molecules on antigen-presenting cells (APCs) and transduces a negative signal that inhibits stimulation of an immune response. LILRB3 may control inflammatory responses and cytotoxicity to help focus the immune response and limit autoreactivity. LILRB3 becomes activated upon coligation of LILRB3 and immune receptors, such as FCGR2B and the B-cell receptor. LILRB3 may down-regulate antigen-induced B-cell activation by recruiting phosphatases to its immunoreceptor tyrosine-based inhibitor motifs (ITIM).

Construct Details: Full length human LILRB3 gene is subcloned into the Lentiviral expression vector pLTC with an upstream CMV promoter, which can be used for both transient and stable expression in mammalian cells.  It can be co-transfected with the LentiPAK DNA mix (SKU# LP-001) into HEK293 cells to produce high titer lentiviral particles.

The product is shipped at 4°C. Upon receipt, centrifuge the product briefly before opening the vial. It is recommended to store small aliquots at the temperature below –20°C for long-term storage and the product is stable for 6 months.
