Human ADIPOR1/PAQR1/ACDCR1 Gene, Full-length cDNA Clone in Lentivector, Endotoxin-free DNA
ADIPOR1 (adiponectin receptor protein 1) or PAQR1 (progestin and adipoQ receptor family member I) is a member of the ADIPOR family of 7-pass transmembrane (7-TM) proteins. Contrary to G-protein coupled 7-TM receptors, the N-terminus of ADIPOR family member is cytoplasmic and the C-terminus is extracellular. ADIPOR1 and ADIPOR2 acts as receptors for adiponectin, a hormone secreted by adipocytes which regulates fatty acid catabolism and glucose levels. ADIPOR1 may form homooligomers and heterooligomers with ADIPOR2. ADIPOR1 has high affinity for globular adiponectin and low affinity for full-length adiponectin. Binding of adiponectin to ADIPOR1 results in activation of signaling cascade that leads to increased AMPK activity, and ultimately to increased fatty acid oxidation, increased glucose uptake and decreased gluconeogenesis.
Gene Symbol: ADIPOR1; CGI45; PAQR1; ACDCR1; CGI-45; TESBP1A
NCBI Gene ID: 51094
Uniprot Entry: Q96A54
Construct Details: Full length human ADIPOR1 gene is subcloned into the Lentiviral expression vector pLTC with an upstream CMV promoter and with or without a selection marker, which can be used for both transient and stable expression in mammalian cells. It can be co-transfected with the LentiPAK DNA mix (SKU# LP-001) into HEK293 cells to produce high titer lentiviral particles. Multiple choices of a stable antibiotic selection marker are available.
Vector Type: pLTC or pLTC-IRES-Marker (lentiviral expression vector containing a heterologous CMV promoter +/- a selection marker, see the vector map above)
Formulation: Supplied with 10 μg of endotoxin-free plasmid DNA at 200 ng/μl in sterile TE buffer (pH8.0)
Gene Insert Size: 1128 (bp)

Primers: Gene insert coding sequence can be confirmed by following primers:
(a) forward (or 5'-end) primer: 5’-CAAATGGGCGGTAGGCGTG-3’;
(b) reverse (or 3'-end) primer: 5’-CCAGGTTTCCGGGCCCTCAC-3’
Note: The sequence may vary due to the naturally occurring variants or mutations, such as single nucleotide polymorphism, or the artifact during PCR amplification. High fidelity PCR systems should always be used.
