TPBG (trophoblast glycoprotein), also known as oncofetal antigen 5T4 and WAIF1 (wnt-activated inhibitory factor 1), is a single-pass type I transmembrane glycoprotein of the LRR (leucine-rich repeat) protein family. TPBG is an oncofetal antigen that is specific to trophoblast cells except for amniotic epithelium. TPBG is expressed by all types of trophoblasts as early as 9 weeks of development. In adults its expression is limited to a few epithelial cell types but highly abundant in many types of tumor cells. TPBG’s expression is associated with poor clinical outcome in numerous cancers. TPBG contains 1 LRRNT (LRR N-terminal cap), 7 LRR (LRR1-7), and 1 LRRCT (LRR C-terminal cap) domains in the extracellular region. TPBG may function as an inhibitor of Wnt/beta-catenin signaling by indirectly interacting with LRP6 and blocking Wnt3a-dependent LRP6 internalization. The C-terminal part of LRRNT and LRR1 are essential for the inhibition of the Wnt signaling pathway. TPBG may also be involved in cell adhesion and motility.

Construct Details: Full length human TPBG/5T4 gene is subcloned into the Lentiviral expression vector pLTC with an upstream CMV promoter and with or without a selection marker, which can be used for both transient and stable expression in mammalian cells. It can be co-transfected with the LentiPAK DNA mix (SKU# LP-001) into HEK293 cells to produce high titer lentiviral particles. Multiple choices of a stable antibiotic selection marker are available.
