CEA (carcinoembryonic antigen), also kown as CEACAM5, BGP1 and CD66e, is a member of the CEA gene family belonging to the immunoglobulin superfamily (IgSF). CEA family members are either transmembrane or GPI-anchored within two subgroups: the CEA cell adhesion molecules (CEACAM) and the pregnancy-specific glycoproteins.  In human, 7 CEACAMs have been identified including 3 transmembrane proteins (CEACAM1, 3, 4) and 4 GPI-anchored molecules (CEACAM5-8). Human CEA or CEACAM-5 contains 1Ig-like V-type (IgV) domain at the N-terminus, followed by 6 Ig-like C2-type (IgC2) domain and a GPI anchor.  It exists as a homodimer. Intriguingly orthologs of human CEA have not been found in other species. Human CEA is expressed primarily in epithelial cells and functions as an adhesion molecule through homophilic and heterophilic interactions with CEACAM1/CD66a. CEA plays an important role in the development and maintenance of epithelial architecture. CEA is up-regulated in a wide array of human tumors and has been used a s a common cancer biomarker. CEA can promote tumor cell migration, invasion, adhesion, and metastasis.  It may also contribute to tumorigenicity by inhibiting cellular differentiation. GPI-anchored CEA can be released by GPI­PLD, resulting in a soluble molecule that induces tumor metastasis.  CEA has been considered as a a target for immunotherapy.

Construct Details: Expressed as the combination of a heavy chain (HC) containing VH from anti-human CEA mAb and CH1-3 region of human IgG1 and a light chain (LC) encoding VL from anti-human CEA mAb and CL of human lamda 2 light chain.  Exists as a disulfide linked dimer of the HC and LC hetero-dimer under non-reducing condition.

 

Source:  Mamamlian cells stably expressing anti-human CEA human heavy chain and light chain (in human IgG1,L2 format) and growing in chemical-defined media with no animal components or antibiotics 

 

M.W.: 150 (49 kDa for a single HC, 26 kDa for a single LC); Estimated by SDS-PAGE under reducing condition(kDa): 50 kDa + 26 kDa.

 

Purity: >95% judged by SDS-PAGE under reducing condition (see the gel image above, labeled as DTT "+")

 

Formulation: Supplied at 0.5 mg/ml in sterile PBSpH7.4 (carrier free). The purified recombinant monoclonal antibody was labeled with Biotin (3-5 Biotin per IgG) using the standard procedure.

  

Endotoxin Level: <0.1 EU per 1 μg of purified recombinant protein determined by the LAL method

 

Biological Activity: Binds human CEA protein (SKU#FCL2139) with high affinity  (KD < 10 nM) as measured by ELISA.  Specifically recognizes cell surface CEA on human cells by flow cytometry, IHC staining and fluorescent microscopy.

Storage

 

The product is shipped at 4°C. Upon receipt, centrifuge the product briefly before opening the vial. It is recommended to store small aliquots at the temperature below –20°C for long-term storage and the product is stable for 3 months. The undiluted protein can be stored at 4°C for no more than 2 weeks.

 

 

 

Use a manual defrost freezer and avoid repeated freeze-thaw cycles.
