Description

CD146, also known as MUC18 and MCAM, is a single-pass type I transmembrane glycoprotein belonging to the immunoglobulin (Ig) superfamily. CD146 contains 2 Ig-like V-type domains and 3 Ig-like C2-type domains (V-V-C2-C2-C2) in the extracellular region (ECD) in addition to a transmembrane (TM) region and a short cytoplasmic tail. CD146 is expressed in endothelial cells in vascular tissue throughout the body. Its expression appears to be limited to vascular smooth muscle in normal adult tissues. CD146 is involved in cell adhesion, and in cohesion of the endothelial monolayer at intercellular junctions in vascular tissue. CD146 is also an adhesion molecule active in neural crest cells during embryonic development. CD146 is associated with tumor progression and the development of metastasis in human malignant melanoma. Its expression may allow melanoma cells to interact with cellular elements of the vascular system, thereby enhancing hematogeneous tumor spread. CD146 can act as a cell surface receptor that triggers tyrosine phosphorylation of FYN and PTK2/FAK1 as well as a transient increase in the intracellular calcium concentration. CD146 is expressed most strongly on metastatic lesions and advanced primary tumors while it is only rarely detected in benign melanocytic nevi and thin primary melanomas with a low probability of metastasis.  It thus suggests that CD146 may play an important role in tumor progression and metastasis.

Construct Details: Full length human CD146/MUC18/MCAM gene is subcloned into the Lentiviral expression vector pLTC with an upstream CMV promoter and a selection marker, which can be used for both transient and stable expression in mammalian cells.  It can be co-transfected with the LentiPAK DNA mix (SKU# LP-001) into HEK293 cells to produce high titer lentiviral particles.

 

Vector Type: pLTC or pLTC-IRES-Marker (lentiviral expression vector containing a heterologous CMV promoter, see the vector map above)

 

Formulation: Supplied with 10 μg of endotoxin-free plasmid DNA at 200 ng/μl in sterile TE buffer (pH8.0)

Storage

 

The product is shipped at 4°C. Upon receipt, centrifuge the product briefly before opening the vial. It is recommended to store small aliquots at the temperature below –20°C for long-term storage and the product is stable for 6 months. 

 

 

Use a manual defrost freezer and avoid repeated freeze-thaw cycles.
