Connective Tissue Growth Factor (CTGF) is a member of the CCN (CYR61/CTGF/NOV) family of secreted proteins that share a multimodular organization. Each CCN member contains an IGF­binding protein domain, a von Willebrand factor type C domain, a thrombospondin type I domain, and a cysteine knot domain. The multimodular CTGF binds multiple ligands and shows effects in development, differentiation, and tumorigenesis. CTGF is a mitogen that is secreted by vascular endothelial cells. It plays a role in chondrocyte proliferation and differentiation, cell adhesion in many cell types, and is related to platelet-derived growth factor. The C­terminal cysteine knot motif contains the heparin and low-density lipoprotein receptor (LDLR) binding sites that contributes to the adhesive and mitogenic properties of CTGF.  Certain polymorphisms in this gene have been linked with a higher incidence of systemic sclerosis. CTGF is the central mediator of tissue remodeling and fibrosis.  It is implicated in tumor progression and metastasis.  It promotes epithelial to mesenchymal transition (EMT), a process whereby normal epithelial cells become migratory, matrix-producing fibroblasts important for invasion and metastasis.  Elevated CTGF levels are found in desmoplastic cancer where it supports the formation of the extensive fibrous connective tissue (desmoplasia) around cancer.  Blockade of the expression or activity of CTGF reduces tumor growth and metastasis in serveral cancer animal model.

Construct Details: Expressed as the combination of a heavy chain (HC) containing VH from anti-CTGF mAb and CH1-3 region of human IgG1 and a light chain (LC) encoding VL from anti-CTGF mAb and CL of human kappa light chain.  Exists as a disulfide linked dimer of the HC and LC hetero-dimer under non-reducing condition (see the gel image above, labeled as DTT "-").

 

Source: Mammalian cells stably expressing anti-CTGF human IgG1 HC and LC and growing in chemical-defined media with no animal component or antibiotics.

Storage

 

The product is shipped at 4°C. Upon receipt, centrifuge the product briefly before opening the vial. It is recommended to store small aliquots at the temperature below –20°C for long-term storage and the product is stable for 3 months. The undiluted protein can be stored at 4°C for no more than 2 weeks.

 

Use a manual defrost freezer and avoid repeated freeze-thaw cycles.
