VEGF, also known as VEGF-A (vascular endothelial growth factor A), VPF (vascular permeability factor) or MVCD1 (microvascular complications of diabetes 1), is the founding member of the VEGF family, including VEGF-A, VEGF-B, VEGF-C, VEGF-D, VEGF-E, and PIGF. VEGFs are secreted polypeptides with a highly conserved receptor-binding cystine-knot structure similar to that of the platelet-derived growth factors (PDGF). VEGF plays important roles in vascular development and in diseases involving abnormal growth of blood vessels such as tumor-related angiogenesis. VEGF is a growth factor active in angiogenesis, vasculogenesis and endothelial cell growth. It induces endothelial cell proliferation, promotes cell migration, inhibits apoptosis and increases permeabilization of blood vessels. VEGF is known to bind to the FLT1 (VEGFR1) and KDR (VEGFR2) receptors, heparan sulfate and heparin. Targeting VEGF and its signaling pathway has shown therapeutic benefits in multiple cancer types. The purified recombinant monoclonal antibody binds both human and mouse VEGF with high affinity. It potently blocks VEGF signaling activity. It also inhibits HUVEC proliferation and tumor cell growth.
Gene Symbol: VEGFA; VEGF-A; VPF; MVCD1
NCBI Gene ID: 7422 (human), 22339 (mouse)
Uniprot Entry: P15692 (human), Q00731 (mouse)
Construct Details: Expressed as the combination of a heavy chain (HC) containing VH from anti-h,mVEGF mAb and CH1-3 region of human IgG1 and a light chain (LC) encoding VL from anti-h,mVEGF mAb and CL of human kappa light chain. Exists as a disulfide linked dimer of the HC and LC hetero-dimer under non-reducing condition (see the gel image above, labeled as DTT "-").
Source: Mamamlian cells stably expressing anti-h,mVEGF human heavy chain and light chain and growing in chemical-defined media with no animal components or antibiotics
M.W.: Calculated molecular mass (kDa): 144.8 (49.2 kDa for a single HC, 23.2 kDa for a single LC); Estimated by SDS-PAGE under non-reducing condition(kDa): 150
Purity: >95% judged by SDS-PAGE under reducing condition (see the gel image above, labeled as DTT "+")
Formulation: Supplied at 1.0 mg/ml in sterile PBS pH7.4 (carrier free).
Endotoxin Level: <0.1 EU per 1 μg of purified recombinant protein determined by the LAL method
Biological Activity: The recombinant antibody binds both human and mouse VEGF (SKU#FCL0387, F#CL0388, #FCL0406, #FCL0407) with equally high affinity (KD < 10 nM as measured by ELISA). It blocks VEGF signaling activity. It also inhibits HUVEC proliferation and tumor cell growth. The recombinant antibody has the same binding specificity as anti-h,mVEGF mIgG2a (SKU#MAB0337).
