Description

TREM2 (Triggering receptor expressed on myeloid cells 2)  is a single pass type I transmembrane glycoprotein that belongs to the Immunoglobulin superfamily (IgSF) with 1 Ig-like V-type domain in the extracellular region.  TREM2 forms a receptor signaling complex with the TYROBP (TYRO protein tyrosine kinase binding protein), which mediates signaling and cell activation following ligand binding.  TREM2 triggers activation of the immune responses in macrophages and dendritic cell to produce constitutive inflammatory cytokines. TREM2 also acts as a receptor for amyloid-beta protein 42 and mediates its uptake and degradation by microglia.   Binding to amyloid-beta 42 mediates microglial activation, proliferation, migration, apoptosis and expression of pro-inflammatory cytokines.  TREM2 is  required for microglial phagocytosis of apoptotic neurons and myelin debris after neuronal injury.  TREM2 also acts as a receptor for lipoprotein particles and for apolipoproteins.  Defects in the TREM2 gene are a cause of polycystic lipomembranous osteodysplasia with sclerosing leukoencephalopathy 2 (PLOSL2), an autosomal recessive disease characterized by presenile frontal dementia.

Construct Details: Full length human TREM2 gene is subcloned into the Lentiviral expression vector pLTC with an upstream CMV promoter and with or without a selection marker, which can be used for both transient and stable expression in mammalian cells.  It can be co-transfected with the LentiPAK DNA mix (SKU# LP-001) into HEK293 cells to produce high titer lentiviral particles. Multiple choices of a stable antibiotic selection marker are available.

Storage

 

The product is shipped at 4°C. Upon receipt, centrifuge the product briefly before opening the vial. It is recommended to store small aliquots at the temperature below –20°C for long-term storage and the product is stable for 6 months. 

 

 

Use a manual defrost freezer and avoid repeated freeze-thaw cycles.
