The PRNP prion protein is a glycosylphosphatidylinositol (GPI)-anchored membrane glycoprotein of the prion family. PRNP has a tendency to aggregate into rod-like structures. PRNP contains a highly unstable region of 5 tandem octapeptide repeats. Mutations in the repeat region as well as elsewhere in the PNRP gene have been associated with Creutzfeldt-Jakob disease, fatal familial insomnia, Gerstmann-Straussler disease, Huntington disease-like 1, and kuru. PRNP’s primary physiological function is unclear. May play a role in neuronal development and synaptic plasticity. PNRP may be required for neuronal myelin sheath maintenance. PRNP may promote myelin homeostasis through acting as a agonist for ADGRG6 receptor. It may also play a role in iron uptake and iron homeostasis. Soluble oligomers of PNRP are toxic to cultured neuroblastoma cells and induce apoptosis. PNRP also provides Cu2+ or ZN2+ for the ascorbate-mediated GPC1 deaminase degradation of its heparan sulfate side chains. An overlapping open reading frame has been found for this gene that encodes a smaller, structurally unrelated protein, AltPrp, which has no apparent direct functional relation to PNRP since a mutation that removes the start codon of the AltPrP has no apparent effect on the biology of PRNP.

Construct Details: Full length human PRNP/CD230 gene is subcloned into the Lentiviral expression vector pLTC with an upstream CMV promoter and with or without an antibiotic selection marker, which can be used for both transient and stable expression in mammalian cells. It can be co-transfected with the LentiPAK DNA mix (SKU# LP-001) into HEK293 cells to produce high titer lentiviral particles. Multiple choices of a stable antibiotic selection marker are available.
