Description

DDR2 (epithelial discoidin domain receptor 2) receptor tyrosine kinase (RTK), is a single-pass type I transmembrane glycoprotein that belongs to the insulin receptor (InsR)subfamily of the tyrosine protein kinase family. DDR2 comprises a 378-aa extracellular domain (ECD), a transmembrane region (TM), and a 434-aa intracellular kinase domain.  DDR2 may play a key role in the communication of cells with their microenvironment and be involved in the regulation of cell growth, differentiation and metabolism. Like other RTKs, DDR2 transduces signals across the membrane has been shown to be ligand induced receptor oligomerization and subsequent intracellular phosphorylation. This autophosphorylation leads to phosphorylation of cytosolic targets as well as association with other molecules, which are involved in pleiotropic effects of signal transduction.  DDR2 contains a distinct extracellular region encompassing a factor VIII-like domain.  DDR2 is required for normal bone development and regulates osteoblast differentiation and chondrocyte maturation via a signaling pathway that involves MAP kinases and leads to the activation of the transcription factor RUNX2. DDR2 regulates remodeling of the extracellular matrix by up-regulation of the collagenases MMP1, MMP2 and MMP13, and thereby facilitates cell migration and tumor cell invasion. DDR2 promotes fibroblast migration and proliferation, and thereby contributes to cutaneous wound healing.  Mutations in DDR2 are associated with Spondyloepimetaphyseal dysplasia short limb-hand type (SEMD-SL).

Construct Details: Full length human DDR2 gene is subcloned into the Lentiviral expression vector pLTC with an upstream CMV promoter and with or without an antibiotic selection marker, which can be used for both transient and stable expression in mammalian cells.  It can be co-transfected with the LentiPAK DNA mix (SKU# LP-001) into HEK293 cells to produce high titer lentiviral particles. Multiple choices of a stable antibiotic selection marker are available.

Storage

 

The product is shipped at 4°C. Upon receipt, centrifuge the product briefly before opening the vial. It is recommended to store small aliquots at the temperature below –20°C for long-term storage and the product is stable for 6 months. 

 

 

Use a manual defrost freezer and avoid repeated freeze-thaw cycles.
