Description

Cadherins are calcium-dependent cell adhesion proteins that preferentially interact with themselves in a homophilic manner in connecting cells.  Cadherins thus contribute to the sorting of heterogeneous cell types.  Cadherin12 or CDH12 is a 794 amino acid (aa) single pass type I transmembrane protein of the cadherin superfamily.  CDH12 contains an N-terminal signal peptide, a 555-aa extracellular domain (ECD) with 5 cadherin domains and 4 potential glycosylation sites, a transmembrane domain (TM), and a 157-aa highly conserved cytoplasmic tail.  CDH12 is a type II cadherin that may play an important role in cell differentiation and morphogenesis. Type II (atypical) cadherins are defined based on their lack of a histidine-alanine-valine (HAV) cell adhesion recognition sequence specific to type I cadherins. CDH12 is expressed specifically in the brain and its temporal pattern of expression suggests a role during a critical period of neuronal development, perhaps specifically during synaptogenesis.

Construct Details: Full length human CDH12 gene is subcloned into the Lentiviral expression vector pLTC with an upstream CMV promoter and with or without a selection marker, which can be used for both transient and stable expression in mammalian cells.  It can be co-transfected with the LentiPAK DNA mix (SKU# LP-001) into HEK293 cells to produce high titer lentiviral particles. Multiple choices of a stable antibiotic selection marker are available.

Storage

 

The product is shipped at 4°C. Upon receipt, centrifuge the product briefly before opening the vial. It is recommended to store small aliquots at the temperature below –20°C for long-term storage and the product is stable for 6 months. 

 

 

Use a manual defrost freezer and avoid repeated freeze-thaw cycles.
