Cadherins are calcium-dependent cell adhesion proteins that preferentially interact with themselves in a homophilic manner in connecting cells. Cadherins thus contribute to the sorting of heterogeneous cell types. Cadherin-1 or CDH1 is an 882 amino acid (aa) single pass type I transmembrane protein of the cadherin superfamily. CDH1 contains an N-terminal signal peptide, a 555-aa extracellular domain (ECD) with 5 cadherin domains and 4 potential glycosylation sites, a transmembrane domain (TM), and a 152-aa highly conserved cytoplasmic tail. CDH1 is a disulfide-linked homodimer and a component of the E-cadherin/catenin adhesion complex composed of at CDH1, beta-catenin/CTNNB1 or gamma-catenin/JUP, and alpha-catenin/CTNNA1. The ECD of CDH1 mediates bacterial adhesion to mammalian cells and the cytoplasmic domain is required for internalization. CDH1 is involved in mechanisms regulating cell-cell adhesions, mobility and proliferation of epithelial cells. CDH1 plays a potent invasive suppressor role and acts as a ligand for integrin alpha-E/beta-7. Mutations in the CDH1 gene are correlated with gastric, breast, colorectal, thyroid and ovarian cancer. Loss of function of the CDH1 gene may contribute to cancer progression by increasing proliferation, invasion, and/or metastasis.
Gene Symbol: CDH1; CD324; UVO; CDHE; ECAD; E-cadherin; LCAM; Arc-1; Cadherin-1
NCBI Gene ID: 999
Uniprot Entry: P12830
Construct Details: Full length human CDH1/CD324 gene is subcloned into the Lentiviral expression vector pLTC with an upstream CMV promoter and with or without a selection marker, which can be used for both transient and stable expression in mammalian cells. It can be co-transfected with the LentiPAK DNA mix (SKU# LP-001) into HEK293 cells to produce high titer lentiviral particles. Multiple choices of a stable antibiotic selection marker are available.
Vector Type: pLTC or pLTC-IRES-Marker (lentiviral expression vector containing a heterologous CMV promoter +/- a selection marker, see the vector map above)
Formulation: Supplied with 10 μg of endotoxin-free plasmid DNA at 200 ng/μl in sterile TE buffer (pH8.0)
Gene Insert Size: 2649 (bp)

Primers: Gene insert coding sequence can be confirmed by following primers:
(a) forward (or 5'-end) primer: 5’-CAAATGGGCGGTAGGCGTG-3’;
(b) reverse (or 3'-end) primer: 5’-CCAGGTTTCCGGGCCCTCAC-3’
Note: The sequence may vary due to the naturally occurring variants or mutations, such as single nucleotide polymorphism, or the artifact during PCR amplification. High fidelity PCR systems should always be used.
