IL12RB2 (interleukin-31 receptor subunit beta 2) is a single pass type I transmembrane glycoprotein of the type I cytokine receptor family, type 2 subfamily. IL12RB2 consists of an extracellular region with 5 fibronectin type III (FN3) domains, and a juxtamembrane WSXWS motif, a transmembrane domain (TM), and a cytoplasmic tail with a box 1 motif. The WSXWS motif is necessary for proper protein folding and efficient intracellular transport and cell-surface receptor binding, while the box 1 motif is required for JAK interaction and activation. IL12RB2 is a subunit of the IL12 receptor complex with IL12RB1/CD212. The co-expression of IL12RB2 and IL12RB1 leads to the formation of high-affinity IL12 binding sites and reconstitution of IL12 dependent signaling. The IL12RB2 subunit is the signaling component coupling to the JAK2/STAT4 pathway to promote the proliferation of T-cells as well as NK cells. The expression of IL12RB2 is up-regulated by interferon gamma in Th1 cells, and plays a role in Th1 cell differentiation. The up-regulation of the IL12RB2 gene is found to be associated with a number of infectious diseases, such as Crohn's disease and leprosy, which is thought to contribute to the inflammatory response and host defense.

Construct Details: Full length human IL12RB2 gene is subcloned into the Lentiviral expression vector pLTC with an upstream CMV promoter and with or without a selection marker, which can be used for both transient and stable expression in mammalian cells. It can be co-transfected with the LentiPAK DNA mix (SKU# LP-001) into HEK293 cells to produce high titer lentiviral particles. Multiple choices of a stable antibiotic selection marker are available.
