Description

CD212 or IL12RB1 (interleukin-31 receptor subunit beta 1) is a single pass type I transmembrane glycoprotein of the type I cytokine receptor family, type 2 subfamily.  IL12RB1/CD212 consists of an extracellular region with 5 fibronectin type III (FN3) domains, and a juxtamembrane WSXWS motif, a transmembrane domain, and a cytoplasmic tail with a box 1 motif.  The WSXWS motif is necessary for proper protein folding and efficient intracellular transport and cell-surface receptor binding, while the box 1 motif is required for JAK interaction and activation.  IL12RB1/CD212 binds to IL12 with a low affinity, and is thought to be a part of IL12 receptor (IL12R) complex. IL12RB1/CD212 forms a disulfide-linked oligomer, which is required for its IL12 binding activity. The co-expression of IL12RB1 and IL12RB2 can lead to the formation of high-affinity IL12 binding sites and reconstitution of IL12 dependent signaling. IL12RB1 also associates also with IL23R to form a receptor complex to transduce IL23 signaling probably through activation of the Jak-Stat cascade. Mutations in the IL12RB1 gene impair the development of IL17-producing T lymphocytes and result in increased susceptibility to mycobacterial infections, resulting in a condition termed immunodeficiency 30 (IMD30).

Construct Details: Full length human IL12RB1/CD212 gene is subcloned into the Lentiviral expression vector pLTC with an upstream CMV promoter and with or without a selection marker, which can be used for both transient and stable expression in mammalian cells.  It can be co-transfected with the LentiPAK DNA mix (SKU# LP-001) into HEK293 cells to produce high titer lentiviral particles. Multiple choices of a stable antibiotic selection marker are available.

Storage

 

The product is shipped at 4°C. Upon receipt, centrifuge the product briefly before opening the vial. It is recommended to store small aliquots at the temperature below –20°C for long-term storage and the product is stable for 6 months. 

 

 

Use a manual defrost freezer and avoid repeated freeze-thaw cycles.
