CD118, also known as LIFR (leukemia inhibitory factor receptor), is a single pass type I transmembrane glycoprotein of the type I cytokine receptor family, type 2 subfamily.  CD118/LIFR consists of an extracellular region with 6 fibronectin type III (FN3) domains and a juxtamembrane WSXWS motif, a transmembrane domain, and a cytoplasmic tail with a box 1 motif.  The WSXWS motif is necessary for proper protein folding and efficient intracellular transport and cell-surface receptor binding, while the box 1 motif is required for JAK interaction and activation. LIFR/CD118 combines with a high-affinity converter subunit, IL6ST/gp130, to form a receptor complex that mediates the action of LIF, a pleitropic cytokine that is involved in cellular differentiation, proliferation and survival in the adult and the embryo. LIFR/CD118 is the signal-transducing molecule in the complex with IL6ST. Many cytokines including IL6, LIF, OSM, CNTF, IL11 and CT1 utilize IL6ST/gp130 for initiating signal transmission. Mutations in the LIFR/CD118 gene cause Schwartz-Jampel syndrome type 2 (SJS2), a disease belonging to the group of the bent-bone dysplasias. A translocation of the LIFR/CD118 gene is associated with salivary gland pleiomorphic adenoma, a common type of benign epithelial tumor of the salivary gland. The soluble form of LIFR/CD118 inhibits the biological activity of LIF by blocking its binding to receptors on target cells.

Construct Details: Full length human LIFR/CD118 gene is subcloned into the Lentiviral expression vector pLTC with an upstream CMV promoter and with or without a selection marker, which can be used for both transient and stable expression in mammalian cells.  It can be co-transfected with the LentiPAK DNA mix (SKU# LP-001) into HEK293 cells to produce high titer lentiviral particles. Multiple choices of a stable antibiotic selection marker are available.

Storage

 

The product is shipped at 4°C. Upon receipt, centrifuge the product briefly before opening the vial. It is recommended to store small aliquots at the temperature below –20°C for long-term storage and the product is stable for 6 months. 

 

 

Use a manual defrost freezer and avoid repeated freeze-thaw cycles.
