Description

CD239, also known as BCAM (basal cell adhesion molecule) and Lu (Lutheran blood group glycoprotein), is a single pass type I transmembrane proteins of the Ig superfamily (IgSF). There are two alternatively spliced variants of CD239, which similar structures except for the cytoplasmic tails (one lacking a 40 amino acid SH3 motif). A polymorphism at position 77 within the extracellular domain (ECD) is the basis for the difference between the Lua and Lub Lutheran blood groups.  CD239 contains 5 Ig-like (2 V-type and 3 C2-type) motifs in the extracellular domain (ECD).  CD239 is widely expressed in epithelial and endothelial tissues. It is also expressed on vascular and visceral smooth muscle cells and at the neuromuscular junction of skeletal muscle. CD239 shares a significant sequence similarity with the CD146/MUC18 and CD166/ALCAM. CD239 cooperates with Integrins β1 and αVβ3 as an adhesion receptor for Laminins. Laminins are found in all basement membranes and are involved in cell differentiation, adhesion, migration, and proliferation.  CD239 is upregulated on carcinomas (particularly ovarian), sarcomas, astrocytomas, and melanomas. In addition CD239 interacts with integrin in sickle red cells, and thus may potentially play a role in vaso-occlusive episodes. Polymorphisms in this gene define some of the antigens in the Lutheran system and also the Auberger system. Inactivating variants of this gene result in the recessive Lutheran null phenotype, Lu(a-b-), of the Lutheran blood group.

Construct Details: Full length human CD239/BCAM gene (encoding UniProt#P50895) is subcloned into the Lentiviral expression vector pLTC with an upstream CMV promoter and with or without a selection marker, which can be used for both transient and stable expression in mammalian cells.  It can be co-transfected with the LentiPAK DNA mix (SKU# LP-001) into HEK293 cells to produce high titer lentiviral particles. Multiple choices of a stable antibiotic selection marker are available.

Storage

 

The product is shipped at 4°C. Upon receipt, centrifuge the product briefly before opening the vial. It is recommended to store small aliquots at the temperature below –20°C for long-term storage and the product is stable for 6 months. 

 

 

Use a manual defrost freezer and avoid repeated freeze-thaw cycles.
