Description

CD66a, also known as CEACAM1 (carcinoembryonic antigen (CEA)-related cell adhesion molecule 1) and BGP (biliary glycoprotein), is a single pass type I tranmembrane protein of the immunoglobulin superfamily (IgSF). CEA family members are either transmembrane or GPI­-anchored within two subgroups: the CEACAMs and the pregnancy-specific glycoproteins.  Members of the CEA family consist of a single N domain, with structural similarity to the Ig variable (IgV) domains, followed by a variable number of Ig constant-like 1 (IgC1) and/or 2 (IgC2) domains.  In human, 7 CEACAMs have been identified including 3 transmembrane proteins (CEACAM1, 3, 4) and 4 GPI-anchored molecules (CEACAM5-8). Human CEACAM1 contains 1 Ig-like V-type (IgV) motif and 3 Ig-like C2-type (IgC2) motifs in the N-terminal extracellular domain (ECD), followed by a transmembrane domain (TM) and an ITIM motif and a calmodulin binding site in the cytoplasmic region. CEACAM1 has 3 soluble and 7 transmembrane isoforms due to alternate splicing. The functions of these isoforms are not clear. CEACAM1 is an adhesion molecule mediating cell adhesion via both homotypic and heterotypic associations with CEACAM5/CEA and CEACAM6/NCA.  CEACAM1 is highly glycosylated and can be detected on leukocytes, epithelia, and endothelia. CEACAM1 is involved in multiple cellular processes including differentiation, angiogenesis, apoptosis, metastasis, as well as the modulation of innate and adaptive immune responses.

Construct Details: Full length human CEACAM1 gene is subcloned into the Lentiviral expression vector pLTC with an upstream CMV promoter and with or without a selection marker, which can be used for both transient and stable expression in mammalian cells.  It can be co-transfected with the LentiPAK DNA mix (SKU# LP-001) into HEK293 cells to produce high titer lentiviral particles. Multiple choices of a stable antibiotic selection marker are available.

Storage

 

The product is shipped at 4°C. Upon receipt, centrifuge the product briefly before opening the vial. It is recommended to store small aliquots at the temperature below –20°C for long-term storage and the product is stable for 6 months. 

 

 

Use a manual defrost freezer and avoid repeated freeze-thaw cycles.
